ABSTRACT

Six compounds were isolated from the aerial part of cultivated Clerodendranthus spicatus in Hainan with various chromatographic techniques, and their structures were determined as:1-dehydroxy-1-oxo-rupestrinol(1),N-trans-feruloyltyramine(2),methyl 3,4-dihydroxyphenyllactate(3),caffein acid(4),methyl caffeate(5) and ethyl caffeate(6),via analysis of physicochemical properties and spectroscopic evidence.Compound 1 was a new compound, while compounds 2 and 3 were isolated from C.spicatus for the first time.Biological activity results showed that compounds 2-4 exhibited α-glucosidase inhibitory activity with different inhibition ratio.

ABSTRACT

Objective: To investigate the water-soluble phenolic acids from Clerodendranthus spicatus of Dai medicine. Methods: The chemical constituents were separated and purified by D101, Sephadex LH-20, MCI, and preparative HPLC column chromatographies. Their structures were determined by physicochemical properties and spectral data analyses. Results: Sixteen constituents were isolated and elucidated as protocatechualdehyde (1), protocatechuic acid (2), 3, 5-dihydroxybenzaldehyde (3), 4-hydroxybenzoic acid (4), 4-hydroxybenzaldehyde (5), vanillic acid (6), danshensu methyl ester (7), 3, 5-O-dimethyl gallic acid (8), helisterculins A (9), lithospermic acid (10), methyl lithospermate (11), dimethy lithospermate (12), helisterculins B (13), ethyl dihydrocaffeate (14), ferulic acid (15), and dihydroferulic acid (16). Conclusion: Compounds 3, 6-9, and 13-16 are obtained from Clerodendranthus Kudo for the first time.

ABSTRACT

Objective To optimize the extraction process of active phenols from Clerodendranthus spicatus (Thunb) C.Y.Wu (CS). Methods The contents of total phenols, rosmarinic acid, caffeic acid, eupatorin and the yield of extract were chosen as indexes. Five methods, including methods of alkaline water extraction, alkaline water extraction with borax, water decoction, warmimpregnation with water and reflux with alcohol were compared. The extracting conditions were optimized by the orthogonal test and the single factor experiment. Results Reflux with alcohol was found to be the best way for phenols from CS. The optimized process was selected. CS was extracted with 50% alcohol twice. The extraction time of each time was 1.5 h. The volume of 50% alcohol was 10 times that of the raw material. Conclusion The optimum process is reasonable, stable and practicable.

ABSTRACT

OBJECTIVE: To study the phenolic acids from the aerial part of Clerodendranthus spicatus. METHODS: The phenolic acids were isolated from the aqueous extract of C. spicatus by various chromatographic methods, and their structures were elucida-ted on the basis of their physicochemistry properties and spectral data. RESULTS: A new phenolic acid possess a unique bicycle [2.2.2] octane skeleton was obtained from the aqueous extract of the aerial part of C. spicatus. CONCLUSION: Compound 1 is a new phenolic acid and named cleroden E.

ABSTRACT

Objective To choose an appropriate coloured system and develop a method for the determination of the main phenolic in Dai Medicine -Clerodendranthus spicatus from different areas. Methods The main phenolic in Clerodendranthus spicatus from 19 areas was determined at 500 nm wavelength, with rosmarinic acid as reference and sodium nitrite-aluminum nitrate-sodium hydroxide as colour-developing system. Results Rosmarinic acid presented good linear relationship (r2=0.9993) in the concentration range of 8.792-17.584μg/ml.The average recovery was 100.73%and RSD was 0.85%.The content of the main phenolic in Clerodendranthus spicatus ranged from 1.16 % to 3.49 %. Conclusion The method is appropriate for the determination of the main phenolic in Clerodendranthus spicatus. The main phenolic content in samples from Yunnan province is relatively high.
